What is this tube with the chrome cap on the manifold for? It looks like a breather tube maybe?
Is this hole in the back of the manifold supposed to be there?

That looks like the oil fill tube to me, but I have been wrong before!
When in doubt, hit the gas. It may not be the right answer, but at least it ends the suspense!!

Old style and some aftermarket manifolds had to have an oil fill tube at the front. The rear is the old road draft tube opening. Fill the rear with a freeze plug.
Old memories of the only 332 I ever saw and 352s before 59.

It is the oil fill / breather. My 64 wagon came that way from the factory. The valve covers do not have any holes on 64
and earlier FE's and are nicknamed "baldies". If your engine has an oil fill hole on the valve covers you can remove it and use
a freeze/core plug to block it off. The hole in the back is for the pcv valve or road draft tube.
I see what looks like a PCV valve on the right side valve cover so I guess somebody just found an old intake to run. I would still block it off and make sure there is a vent style cap on the left side
